چکیده

We explored photothermal quantitative phase imaging (PTQPI) of living cells with functionalized nanoparticles (NPs) utilizing a cost-efficient setup based on a cell culture microscope. The excitation light was modulated by a mechanical chopper wheel with low frequencies. Quantitative phase imaging (QPI) was performed with Michelson interferometerbased off-axis digital holographic microscopy and a standard industrial camera. We present results from PTQPI observations on breast cancer cells that were incubated with functionalized gold NPs binding to the epidermal growth factor receptor. Moreover, QPI was used to quantify the impact of the NPs and the low frequency light excitation on cell morphology and viability.

Dynamic measurements of flowing cells labeled by gold nanoparticles using full-field photothermal interferometric imaging.

We present highly dynamic photothermal interferometric phase microscopy for quantitative, selective contrast imaging of live cells during flow. Gold nanoparticles can be biofunctionalized to bind to specific cells, and stimulated for local temperature increase due to plasmon resonance, causing a rapid change of the optical phase. These phase changes can be recorded by interferometric phase micr...

Fast wide-field photothermal and quantitative phase cell imaging with optical lock-in detection

We present a fast, wide-field holography system for detecting photothermally excited gold nanospheres with combined quantitative phase imaging. An interferometric photothermal optical lock-in approach (POLI) is shown to improve SNR for detecting nanoparticles (NPs) on multiple substrates, including a monolayer of NPs on a silanized coverslip, and NPs bound to live cells. Furthermore, the set up...

Prediction of photothermal phase signatures from arbitrary plasmonic nanoparticles and experimental verification

We present a new approach for predicting spatial phase signals originated from photothermally excited metallic nanoparticles of arbitrary shapes and sizes. Heat emitted from the nanoparticle affects the measured phase signal via both the nanoparticle surrounding refractive index and thickness changes. Since these particles can be bio-functionalized to bind certain biological cell components, th...
